Use of Unpollinated Ovaries for the Multiplication of TGMS Lines for Hybrid Rice

Genetic Material Used

VN-01, 162, and the new high-yielding line 271-263, were used as male sterile lines of the TGMS type. CR 203 cv was used as the control.

Three Media Tested

Of the 3 kinds of media tested, MS 1 (Kinetin 0.2mg/l + IAA 0.2mg/l), MS 2 (Kinetin 0.2 mg/l) and MS 3 (Kinetin 0.2 mg/l + NAA 1mg/l), MS 1 was the best for callus induction (induction rate 95%).

Timing of Sample Collection

Callus induction was most successful when samples were collected at flower phase 1 and 2 (the length of the rice flower is about 5-10, 15-20 mm). Three combinations of phytohormones were used to regenerate plants from callus. These were MS 4 (NAA 1mg/l + Kinetin 4mg/l), MS 5 (NAA 1mg/l + Kinetin 2mg/l + BAP 0.25 mg/l), MS 6 (NAA 1 mg/l + BAP 2 mg/l)

Rooting Percentage

Regenerated plants easily rooted in the medium of LS with the addition of a small quantitiy of NAA. The rooting percentage for CR 203 was 94.7 %, while that for VN-01 was 96.7% .
